ABSTRACT

BACKGROUND: The testes are highly susceptible to the adverse effects of chemotherapy and radiation at all stages of life. Exposure to these threats mainly occurs during cancer treatment and as an occupational hazard in radiation centers. The present study investigated the regenerative ability of adipose-derived mesenchymal stem cells (ADMSCs) against the adverse effects of cisplatin on the structure and function of the testes. METHODS: New Zealand white rabbits (N = 15) were divided into three groups of five: a negative control group (no treatment), a cisplatin group (single dose of cisplatin into each testis followed three days later by a PBS injection), and a cisplatin + ADMSCs group (cisplatin injection followed three days later by an ADMSC injection). On day 45 post-treatment, serum testosterone levels were evaluated, and the testes and epididymis were collected for histology, oxidative stress examination, and epididymal sperm analysis. RESULTS: Cisplatin caused damage to the testicular tissue and decreased serum testosterone levels, epididymal sperm counts, and oxidants. An antioxidant imbalance was detected due to increasing malondialdehyde (MDA) and reduced glutathione (GSH) levels in testicular tissue. The ADMSC-treated group displayed a moderate epididymal sperm count, adequate antioxidant protection, suitable hormone levels, and enhanced testicular tissue morphology. CONCLUSIONS: ADMSCs treatment repaired damaged testicular tissue, enhanced biochemical parameters, and modified pathological changes caused by cisplatin.

ABSTRACT

Non-strangulated acquired hernias in stallions are rare, especially when the herniated content is not intestinal loops. Thus, the aim of the current study is to describe a case of acquired non-strangulated inguinoscrotal hernia in a stallion, whose herniated content was the omentum. The patient was a Criollo stallion with history of rhabdomyolysis, laminitis and bilateral scrotal volume increase observed in the left scrotal region. The animal presented pain in both thoracic limbs, abnormal blood test, especially hypoproteinemia, and mild pain during palpation in the left inguinal ring region. Hydrocele secondary to hypoproteinemia was suspected. After admission, the animal showed signs of acute abdomen, which were clinically reversed. With this, the animal was subjected to ultrasound examination of the scrotal region, whose findings suggested non-strangulated inguinal hernia, although the content could not be identified. Surgical treatment was chosen in order to identify the herniated content and remove the left testicle. Access to the affected scrotum was performed, in which the presence of fluid and a portion of the omentum was observed surrounding the testis and adhering to it. The animal was discharged after he recovered from the surgery and from laminitis. During the breeding season, the stallion remained with a herd of mares for natural mating. After 15 months of surgery, the animal was reassessed and showed no active signs of inflammatory and degenerative processes in the remaining testis. On this occasion, a pregnancy diagnosis was also performed, and all the mares were pregnant. It is concluded that the presence of omentum as a herniated content does not represent a surgical emergency but can make the reproductive prognosis reserved. In addition, removal of the affected testicle can benefit the spermatogenesis of the remaining testicle.(AU)


As hérnias adquiridas não estranguladas em garanhões são raras, principalmente quando o conteúdo herniado não é de alças intestinais. Assim, o objetivo deste trabalho foi descrever um caso de hérnia inguino escrotal adquirida e não estrangulada em garanhão, cujo conteúdo herniado era composto pelo omento. Foi atendido um garanhão da raça Crioula com histórico de rabdomiólise, laminite e aumento de volume escrotal bilateral, evidenciado na região escrotal esquerda. O animal apresentava dor nos membros torácicos, alterações na avaliação sanguínea, destacando-se a hipoproteinemia, e demonstrava dor leve à palpação na região do anel inguinal esquerdo. Suspeitou-se de hidrocele secundária a hipoproteinemia. Após a internação, o animal apresentou sinais de abdômen agudo, revertidos clinicamente. Com isso, realizou-se ultrassonografia da região escrotal, cujos achados sugeriram hérnia inguinal não estrangulada, sem que o conteúdo pudesse ser identificado. Optou-se pelo tratamento cirúrgico, com o intuito de identificação do conteúdo herniado e remoção do testículo esquerdo. Foi realizado acesso à bolsa escrotal afetada, na qual se observou presença de líquido e de uma porção do omento envolvendo o testículo e aderido a ele. O animal recebeu alta após restabelecimento da cirurgia e da laminite. Na propriedade, durante temporada reprodutiva, o garanhão permaneceu com uma manada de éguas para realização de monta natural. Passados 15 meses da cirurgia, o animal foi reavaliado e não demonstrou sinais ativos de processo inflamatório e degenerativos no testículo remanescente. Nessa ocasião, também foi realizado diagnóstico de gestação e todas as éguas encontravam-se prenhes. Conclui-se que a presença de omento como conteúdo herniado não representa uma emergência cirúrgica, mas pode tornar o prognóstico reprodutivo reservado. Ainda, a remoção do testículo afetado pode trazer benefícios à espermatogênese do remanescente.(AU)

ABSTRACT

A cotonicultura tem forte fator de impacto na economia nacional, e o estado de Mato Grosso se destaca por ser o maior produtor de algodão herbáceo e deter o maior rebanho bovino do país, condições essas que estimulam o uso do caroço, da torta e do farelo de algodão na alimentação animal. Considerando que o gossipol está presente nos subprodutos do algodão e que seus efeitos sobre a reprodução podem reduzir a fertilidade dos animais, objetivou-se, com essa pesquisa, avaliar o efeito da ingestão de dietas com diferentes teores de gossipol livre por dia sobre a morfometria testicular e a qualidade seminal de touros da raça Nelore. Foram utilizados 28 touros, distribuídos aleatoriamente em seis tratamentos: T0, 0 grama de gossipol livre/touro/dia ; T1, 1,08 grama de gossipol livre/touro/dia; T2, 2,07 gramas de gossipol livre/touro/dia; T3, 3,24 gramas de gossipol livre/touro/dia; T4, 3,82 gramas de gossipol livre/touro/dia e T5, 5,08 gramas de gossipol livre/touro/dia. Os animais de cada tratamento foram mantidos confinados em área média de 100m2, dotada de bebedouro, cochos para mistura mineral e para volumoso/concentrado. O consumo de 3,24 gramas de gossipol livre/touro/dia alterou a qualidade espermática e a morfometria testicular de touros.(AU)


The cotton industry has a strong impact factor in the Brazilian economy and the state of Mato Grosso stands out for being the largest upland-type cotton producer and also holds the largest cattle herd in the country, conditions that stimulate the use of cottonseed pie and cottonseed meal in animal feed. Whereas gossypol is present in cotton by-products and their effects on reproduction can reduce the animal fertility, this research evaluated the effect of diets with different free gossypol contents in the testicular morphometry and semen quality of Nelore bulls. 28 bulls were randomly distributed in six treatments: T0, receiving no free gossypol; T1, receiving 1.08g / bull / day; T2, receiving 2.07g / bull / day; T3, receiving 3.24g / bull / day; T4, receiving 3.82g / bull / day and T5, receiving 5.08g / bull / day of free gossypol respectively. The animals in each treatment were kept confined in an averaged area of 100m2, having fresh water, troughs for mineral mixture and roughage / concentrate. The consumption of 3.24g of free gossypol / bull / day altered the bulls sperm quality and testicular morphometry.(AU)

ABSTRACT

Due to the doubts and questions about the inflammatory reaction caused by chemical castration, this study aimed to use infrared thermography to detect, evaluate and monitor the inflammatory reaction caused by the intratesticular injection of calcium chloride (CaCl2) 20% with lidocaine 1%. For this, thermographic measurements were taken before (M0), 10 minutes (M1), 1 and 6 hours (M2 and M3), for 7 consecutive days (M4 to M10), at 15 (M11), 30 (M12) and 60 (M13) days after intratesticular injection. Additionally, changes to testicular tissue and effects over spermatogenesis were evaluated by andrological exam before (M0) and 60 days (M13) after intratesticular injection. All cats were orchiectomized at M13, and testicles were submitted to histological analysis. CaCl2 (20%) with lidocaine (1%) administration produced testicular tissue damage and interfered with the spermatogenesis in 70% of treated cats without exacerbating the inflammatory reaction or impairing the cat's welfare. It was concluded that thermographic evaluation is a useful, efficient, easy and quick method to diagnose and monitor cat testicular inflammatory reactions.(AU)


Devido a constantes dúvidas e questionamentos sobre a reação inflamatória ocasionada pela castração química, este estudo objetivou o uso da termografia infravermelha para detectar, avaliar e monitorar a reação inflamatória causada pela injeção intratesticular de cloreto de cálcio (CaCl 2 ) 20% associada à lidocaína 1%. Para isso, medidas termográficas foram aferidas antes (M0), 10 minutos (M1), uma e seis horas (M2 e M3), por sete dias consecutivos (M4 a M10), aos 15 (M11), 30 (M12), e 60 (M13) dias após injeção intratesticular, nos grupos tratado e controle. Todos os gatos foram orquiectomizados no M13, e os testículos submetidos à análise histológica. A injeção CaCl 2 a 20% associada com lidocaína a 1% produziu lesão testicular e interferiu na espermatogênese de 70% dos gatos tratados, sem exacerbar a reação inflamatória ou prejudicar o bem-estar do animal. Foi concluído que a avaliação termográfica é uma ferramenta útil, eficiente, rápida e fácil para o diagnóstico e o monitoramento da reação inflamatória em gatos.(AU)

ABSTRACT

O conhecimento da fisiologia do desenvolvimento testicular e ponderal, a precocidade sexual, a capacidade de produção e qualidade espermática, além dos fatores que potencialmente interferem nestes processos, são importantes para predizer a capacidade reprodutiva dos touros. Entre esses fatores, existem alguns que podem influenciar negativamente na fisiologia reprodutiva do touro, e, dessa forma, reduzir a fertilidade desses animais e causar esterilidade, tais como os fatores diretamente relacionados ao manejo e a nutrição. Dessa forma, torna-se de fundamental importância o conhecimento da fisiologia do desenvolvimento testicular e ponderal, e de como alguns fatores, como o manejo e o clima, podem interferir nestes processos. Portanto, o objetivo desta revisão é demonstrar como alguns fatores do ambiente, tais como o manejo e o clima, podem influenciar nessas características, com vistas à produção animal.(AU)


Knowledge of the physiology and weight of the testicular development, sexual precocity, ability and quality of sperm production, and the factors that potentially interfere with these processes are important for predicting the reproductive capacity of bulls. Among these factors, there are some that can negatively influence their reproductive physiology, and thus reduce their fertility, causing sterility, such as factors directly related to the management and nutrition. Thus, it is of fundamental importance to study the physiology of testicular development and weight, and how certain factors, such as management and climate, can interfere with these processes. Therefore, the aim of this review is to show how some environmental factors, such as management and climate, can influence these characteristics, aiming to animal production.(AU)


El conocimiento de la fisiología del desarrollo testicular y ponderal, la precocidad sexual, la capacidad de producción y la calidad del esperma, además de los factores que potencialmente interfieren en estos procesos, son importantes para predecir la capacidad reproductiva de los toros. Entre estos factores, hay algunos que pueden influir negativamente en la fisiología reproductiva del toro, y por lo tanto reducir la fertilidad de esos animales y causar esterilidad, tales como los factores directamente relacionados al manejo y la nutrición. Por lo tanto, es de fundamental importancia el conocimiento de la fisiología del desarrollo testicular y ponderal, y cómo ciertos factores, como el manejo y el clima, pueden interferir en estos procesos. Así, el objetivo de esta revisión es demostrar cómo algunos factores ambientales, tales como manejo y el clima, pueden influenciar en esas características, con miras a la producción animal.(AU)

ABSTRACT

High temperature is an important factor for reproduction and can induce testicular degeneration. Pentoxifylline is a methylxanthine phosphodiesterase inhibitor with anti - inflammatory and anti - apoptotic properties. Considering the protective propert ies of pentoxifylline and the harmful effects of heat, the present study aimed to use pentoxifylline to reduce the damage induced by heat to the testis. Adult mal e Wistar rats were exposed to testicular heat shock (43ºC for 15 min) , treated with 50 or 100 mg/ k g of pentoxifylline and evaluated at 3, 7, 15, 30 and 60 days after heat shock. Pentoxifylline treatment did not change testicular weight, histomorphometrical parameters or plasma testosterone concentration. However, pentoxifylline inhibited germ cell apoptosis and reduced the severity of pathological lesions at 30 and 60 days after testicular heat shock. In conclusion, pentoxifylline treatment seem ed to inhibit pro - inflammatory and apoptotic mechanisms triggered by testicular heat shock, improving sper matogenesis regeneration.

ABSTRACT

This study was conducted to determine the effect of scrotal insulation and post insulation Proviron® treatment on sperm morphological characteristics and mating behavior. Twelve healthy WAD bucks, free from any clinical or andrological disorder were used. Scrotal insulation was done using insulating bags for 30 days. After insulation, the bucks were divided into two groups. The treatment group received 100mg/head /week of Proviron® for 3 weeks. Semen collection was done using the electroejaculation method in all phases of the study. The proviron® treated bucks, when compared with insulation and post insulation untreated phases, showed significant reduction (p<0.05) in sperm cell abnormalities which reduced from 15.89 +/- 22.89 in post insulation untreated phase to 2.81 +/- 0.83 in post insulation treated phase. The Proviron® treated bucks also showed increased physical vigor by riding, mounting and fighting their untreated counterparts.


Este estudio se realizó para determinar el efecto de aislamiento escrotal y aislamiento posterior al tratamiento con Proviron® sobre las características morfológicas de los espermatozoides y el comportamiento del apareamiento. Fueron utilizadas 12 cabras enanas del Oeste Africano, sanas, libres de cualquier desorden clínico andrológico. El aislamiento escrotal se realizó utilizando bolsas de aislamiento durante 30 días. Después de aislamiento, los machos se dividieron en dos grupos. El grupo de tratamiento recibió 100mg/cada una por semana de Proviron® durante 3 semanas. La recolección de semen se realizó mediante el método de electroeyaculación en todas las fases del estudio. Las cabras tratadas con Proviron®, cuando se compararon con las fases post-aislamiento no tratadas, mostraron una reducción significativa (p<0,05) en las anormalidades de células espermáticas, las cuales se redujeron desde 15,89 +/- 22,89 en las fases post-aislamiento sin tratamiento a 2,81 +/- 0,83 en las fases post-aislamiento tratadas. Las cabras tratadas con Proviron® también mostraron un incremento en el vigor físico, la monta y la lucha contra sus homólogos no tratados.

ABSTRACT

Quatro ovinos machos, com cerca de 12 meses de idade (Ovinos 1-4), foram infectados por via intravenosa com aproximadamente 1,25x10(5) tripomastigotas de Trypanosoma vivax, outros quatro ovinos (Ovinos 5-8) destinaram-se ao grupo controle. Após a infecção, exames clínicos visando avaliar temperatura retal, freqüências cardíaca e respiratória e parasitemia foram realizados diariamente por 30 dias, tempo estabelecido para o término do experimento. A avaliação do hematócrito foi realizada a cada cinco dias. Ao final do período experimental, os animais foram castrados e os testículos e epidídimos submetidos ao exame anatomopatológico. Amostras destes órgãos dos Ovinos 1, 4 e 5 foram tomadas para a realização da reação em cadeia da polimerase (PCR). Os parâmetros clínicos (hipertermia, aumento das freqüências cardíaca e respiratória, aumento de volume dos linfonodos e palidez das mucosas) mantiveram-se para o grupo infectado acima dos valores mostrados pelo grupo controle durante todo o período experimental. A parasitemia foi observada a partir do 3º dia pós-infecção (dpi) com picos nos 6-10os dpi e nos 15-18os dpi. Os Ovinos 1 e 4 apresentaram, a partir do 25º dpi, anemia acentuada. Macroscopicamente, todos os testículos dos animais do grupo infectado apresentaram-se flácidos e com coloração pálida. Microscopicamente, observaram-se degeneração testicular moderada a acentuada, epididimite multifocal e hiperplasia do epitélio epididimário. A análise por PCR de T. vivax nos tecidos testicular e epididimário resultou em 100% de positividade para ovinos infectados experimentalmente. As lesões epididimárias e testiculares associadas à presença do parasita nesses órgãos, detectada por PCR, sugerem a participação do parasita no mecanismo etiopatogênico de danos reprodutivos.(AU)


Four adult sheep (number 1, 2, 3 and 4), all males, were inoculated intravenously with 1ml of blood containing 1.25x10(5) trypomastigotes of Trypanosoma vivax, and Sheep 5, 6, 7 and 8 were used as control. After infection, clinical exams considering rectal temperature, respiratory and cardiac frequencies, and parasitaemia were recorded daily for a 30-day experiment period. Blood samples were obtained for 5-day intervals to hematocrit analysis. At the end of the experimental period, the sheep were orquiectomized. Testes and epididymides from these animals were studied anatomopathologically. Samples from these tissues of Sheep 1, 4 and 5 were taken to polymerase chain reaction (PCR). Clinical parameters remained for the infected group above the values observed in the control group during the experimental period. Parasitaemia was observed on day 3 post-infection, and the highest values occurred between day 6 and 10, and day 15 and 18 post-infection. Sheep 1 and 4 showed severe anemia on day 25 post-infection. All sheep of the infected group showed flabby and palid testes. Histologically, moderate to severe testicular degeneration, multifocal epididymitis and hyperplasia of epididymal epithelium were observed. The result of T. vivax PCR analysis in the testes and epididymal tissues was positive in 100% of the samples of the experimentally infected sheep. Epididymal and testicular lesions associated with the presence of the parasite in these tissues, shown by PCR, suggest the participation of T. vivax in the pathophysiological mechanism of reproductive damage.(AU)

ABSTRACT

Com o objetivo estudar o efeito de dieta contendo gossipol sobre a qualidade seminal e morfologia dos testículos de touros da raça Nelore, utilizaram-se 16 animais, distribuídos em dois tratamentos: T1, oito touros submetidos a dieta contendo 20 por cento de caroço de algodão e T2, oito touros submetidos a dieta isenta de gossipol. Realizaram-se coletas de sêmen para avaliar a qualidade seminal. Os testículos dos animais foram retirados ao final dos 73 dias de experimento, fragmentos dos mesmos foram preparados para serem analisados em microscopia eletrônica de transmissão e outros fixados em lâminas e corados por hematoxilina-eosina. O consumo médio de caroço de algodão/animal/dia de 1,71kg interferiu na morfologia dos testículos; os túbulos seminíferos apresentaram menor espessura de parede, redução no número de camadas de células espermatogênicas e no volume dos núcleos da célula de Sertoli em relação aos do T2. Por microscopia eletrônica observouse que os touros do T1 apresentavam células de Sertoli com membrana citoplasmática enrugada, acúmulo de lipídeos no citoplasma, mitocôndrias deformadas e algumas espermátides estavam em apoptose. Touros do T1 apresentaram redução na motilidade espermática, aumento no percentual de defeitos espermáticos maiores e totais em relação aos animais do T2. O gossipol alterou a morfologia testicular e reduziu a qualidade espermática dos touros [...]


We studied the effect of a gossypol-enriched diet on the quality of semen and the morphology of testes of Nelore bulls. Sixteen animals were scheduled for 2 treatments: T1, 8 bulls fed a diet containing 20 percent cottonseed and T2, 8 bulls fed a gossypol-free diet. Semen samples were collected in order to evaluate the semen quality. At the end of the 73-day period of treatment, the testes were removed and tissue fragments were prepared for transmission electron microscopy or fixed on slides and stained with haematoxylin-eosin for light microscopy. The mean daily intake of cottonseed by T1 bulls was 1.71 kg/animal, which interfered on testes morphology; the seminiferous tubules had less thick walls, fewer layers of the spermatogenic lineage cells and reduced volumes of Sertoli cell nuclei with regard to T2 bulls. Electron microscopy analyses revealed that the Sertoli cells from T1 animals had a rough cytoplasmic membrane, lipid accumulation in the cytoplasm, deformed mitochondriae; in addition, some apoptotic spermatids were seen. T1 bulls showed reduction of sperm motility and an increase of the percentage of major and total sperm defects in relation to the animals of T2 group.
